tribal nations, it’s that offering visitors a variety of options is now necessary for survival. If there’s anything the COVID-19 pandemic has taught tourism-based economies like those of many U.S. Two separately interviewed officials from the tribe’s eight-member legislative government confirmed the cannabis project, but they spoke on condition of anonymity because Mashantucket Pequot leadership had not authorized them to speak publicly. The dispensary, which could open as early as the spring, will be located less than a half-mile from the casino resort-adding to the Mashantucket Pequots’ portfolio of nearby tourist attractions that include a golf course, movie theater, retail shopping mall, trio of spas and dozens of restaurants. The Mashantucket Pequot tribal nation operates Foxwoods Resort Casino on the tribe’s 1,250-acre reservation in the southeast corner of the state. In Connecticut, the tribal owners of one of the East Coast’s most popular casinos also plan to open a mega-dispensary in 2022. Cherokees, Mashantucket Pequots heading the initiativesĪ source close to both initiatives, who asked to remain anonymous due to the sensitivity of the political situation in North Carolina, told Leafly both projects are being planned at mega-dispensary scale, with footprints exceeding 25,000 square feet. But opening a tribal dispensary in a non-medical state like North Carolina would be a precedent-setting action-and it’s unclear whether the Cherokee tribe will wait until the state legalizes medical before opening its dispensary. The Foxwoods-adjacent store may not cause much concern among state or federal officials, because Connecticut is now adult-legal. Earlier this month, the Eastern Band of Cherokee Indians Tribal Council voted to legalize medical marijuana on Cherokee land. In other legal states, tribes typically tax cannabis at a rate comparable to the state, but the tax revenue goes to the tribe, not the state. Native American tribes are not bound by state law, and do not require a state cannabis license. But the North Carolina Compassionate Care Act, a bill to legalize medical cannabis, is currently making progress in the state legislature. The state hasn’t even legalized medical marijuana yet. In North Carolina, all cannabis is strictly prohibited. Retail sales in state-licensed stores are expected to begin in the summer of 2022. On July 1, 2021, it became legal for all adults 21 and older to possess up to 1.5 ounces of marijuana. Connecticut legalized adult-use cannabis earlier this year. This meaning gives a lot of credence to Greek myths that describe Cerberus as a loyal guide for the dead, but a vicious creature who attacks and devours those who attempt to leave the underworld. There have been some ancient sources that have made the claim that Cerberus’s name comes from the Greek word creoboros, which means “flesh-devouring”. Owing to the role that he played in the underworld, Cerberus’s had very sharp hearing it’s been said that he could hear everything that went on around the boundaries of the underworld. And the Latin poet Horace described the beast as having a three-tongued mouth that was poisonous. According to Euphorion, the monster possessed eyes that literally shot out fire. It was often the case that the Greeks believed Cerberus ate raw flesh. However, Quintus Horatius Flaccus, the Roman lyric poet during the era of Emperor Augustus, described Cerberus as having just one dog head and a hundred snake heads. Similarly, ancient Greek lyric poet Pindar of Thebes described Cerberus as a monster with hundred heads. | Image: Heracles, chain in left hand, his club laid aside, calms a two-headed Cerberus, which has a snake protruding from each of his heads, a mane down his necks and back, and a snake tail.Īccording to an account by ancient Greek poet Hesiod, Cerberus possesses a total of fifty heads. In the Greek and Roman art that we have discovered so far, Cerberus is often seen with two visible dog heads. Latin writer and mythographer Fabius Planciades Fulgentius stated that Cerberus’s three heads symbolized three very immense human strife – nature, cause and accident. In some accounts, some authors opined that the heads were symbols of birth, youth and old age. To both ancient Greeks and Romans, Cerberus’s three heads symbolized the past, present and the future. Similarly, his tail was that of a serpent. It was believed that his mane was made from hundreds of poisonous snakes. The ancient Greeks commonly depicted Cerberus as a three-headed hound with razor-sharp teeth and claws similar to that of a lion. However, the common denominator is the terrifying nature of this ancient Greek mythical hellhound. Over the centuries, depictions of Cerberus in art and literature have been diverse. What else was this three-headed monster most known for? In the article below WHE explores all the major myths surrounding Cerberus, a terrifying three-headed dog placed at the entrance of the underworld in Greek mythology. It is for this reason Cerberus was sometimes known as the “hound of Hades”, as the word Hades was also used as a synonym of the underworld in Greek mythology. He was also tasked to perform other duties: first, prevent the dead from escaping the underworld second, stop any living being from making it into the underworld without the permission of Hades. A loyal servant of the Greek god Hades, Cerberus was placed at the gates of hell to gracefully guide the dead into the underworld. In ancient Greek mythology, that role fell to Cerberus, a monstrous three-headed dog whose mane was made from several hundreds of poisonous snakes. In Norse mythology, it is Garmr, the fierce blood-stained wolf/dog. In ancient Egypt, the jackal-headed god Anubis, also known as the god of funerals and embalming, was the one who stood guard at the Underworld. the living, from making their way into the underworld as well as keep the dead from leaving the underworld. The rationale for doing so is to prevent uninvited people, i.e. What is even more interesting is that the place is often guarded by gods and other mythical creatures. A quick look at some of the major mythologies and religions around the world and immediately one cannot help but notice that the concept of the underworld exists in almost every one of them. These alterations increase the affinity for adenosine triphosphate, inducing conformational changes with steric hindrance and interference with the downstream signaling pathway ( 6- 11). Despite these results, after a median period of 10.9 months all patients progress due to the development, in approximately one third of patients, of acquired resistance mutations in the ALK tyrosine kinase domain, such as L1196M, G1269A, C1156Y, L1152R, G1202R, S1206Y, 1151Tins, F1174C, and D1203N. Crizotinib, a I generation multi-TKI, initially developed as a mesenchymal-epithelial transition factor (MET) inhibitor, was the first used in clinic due to results of PROFILE trials showing median progression-free survival (mPFS), objective response rate (ORR) and median duration of response (mDOR) higher than those of chemotherapy in all lines of treatment ( 4, 5). In this setting, the main therapeutic strategy is represented by anti-ALK rearrangement tyrosine kinase inhibitors (TKIs). In approximately 30% of these patients central nervous system (CNS) involvement is present at diagnosis ( 3). Patients affected by this tumor are frequently young, women, with a limited or absent history of smoking and often with a histologic diagnosis of a signet-ring cell adenocarcinoma ( 2). Accepted for publication Sep 18, 2019.Īnaplastic lymphoma kinase (ALK) gene translocations are pro-tumoral driver alterations, present in 3–7% of non-squamous non-small cell lung cancers (NSCLC) ( 1). Keywords: Anti-anaplastic lymphoma kinase tyrosine kinase inhibitors (anti-ALK TKIs) resistance alectinib ceritinib brigatinib In this review we will discuss the most recent results of new TKIs in order to describe a fast growing therapeutic landscape in this setting. Some of them were approved after the failure of crizotinib (ceritinib, alectinib, brigatinib and lorlatinib) and in first line setting (ceritinib, alectinib and brigatinib) while others are still under evaluation for TKI-naive patients such as lorlatinib, ensartinib and entrectinib. For instance, in order to overtake acquired resistance to crizotinib, prolong the control of the disease and manage CNS localizations, several II and III generation TKIs have been developed. This is primarily attributed to poor CNS penetration by crizotinib as many pre-clinical and clinical models suggest. Moreover, 60–90% of patients treated with crizotinib has a progression in the central nervous system (CNS) in absence of extracranial worsening of the disease. Despite its use improved significantly progression-free survival, overall response rate and duration of response of this illness, after a median period of 10.9 months all patients progress due to the development of acquired resistance mutations in the ALK tyrosine kinase domain in approximately one third of patients. The therapeutic strategy depends on anti-ALK tyrosine kinase inhibitors (TKIs) of which crizotinib was the first approved for clinical use.
Side-note for parents: like the first game, this isn't a game for kids as it contains unsettling material, graphic violence, and strong language. But this game didn't make my heart beat as fast as the first game, which I had a crap ton of anxiety when playing. When I played this game and the first one I was connected to a heart rate monitor and had either one of my siblings or my friend record my heart rate. But still, despite its flaws, Outlast 2 is a frightening game that despite its flaws and the fear factor being held back that is an enjoyable experience when playing. The mountain setting and the cult added some sense of dread to the game, but it wasn't as unsettling as the asylum setting and the asylum patient enemies from the first game. The game did give me a lot of scares throughout, but I wasn't traumatized at all from the gameplay, like the first game. While the game was scary, the sequel held back the psychological horror factor unlike the first game where the developers go all out. The answer is no, but outlast two was very damn close to raising the expectations. When I got to play this game at a friends house, I was excited to see if the sequel actually beat the first game. When I played outlast two, the bars were set very high for the sequels to beat. Unforgettable experience, Highly recommended 100%. This game will make you terrorized, stressed out, weirded out, disturbed and maybe traumatized. If you're looking for fun stuff this may not be for you. The graphics and the visuals are insane, these graphics are basically one of the best things I've ever seen in a video game, truly impressive and visually stunning. The setting is on point, both Temple Gate and the School locations are amazingly designed. The soundtrack and the sound effects added more to the experience its eerie aesthetics. The story writing was incredible, extremely complex and very mature in both storytelling and narrative design. Of course the story in the first Outlast is better, the characters at some point are better, but that's not the point. Very effortlessly, Red Barrels just have the power of horror to create a masterful horror story. Outlast II is a bloody, gory, controversial, messed up and disturbed work of pure art. But now I got the chance to play it again, and now I understand it. I played this game for the first time a long time ago, I hated it and left me disappointed, maybe because it didn't live up with the first game. In The Soldier, a monk undergoes a life-changing experience brought about by secret machinations in the far-distant royal court that he knows nothing about. The Mother Provides gives us a woman’s view of the Kingdom’s complex and conflicting religious traditions. The Exiletakes us away from the royal court to examine the daily life of the agricultural peasantry and the unusual family structure that allows them to survive in a brutal environment. While the Kingdom takes pride in its ageless traditions that go back over a thousand years, the ambassador’s very presence marks the beginning of inevitable change and disruption.īeware! takes us back to the days before the investiture and explores the tensions disguised by the elaborate ceremony. In He has returned to us! we are introduced to the Kingdom through the eyes of an ambassador from a neighbouring kingdom as he witnesses the investiture of a new God-King. The language and names are inspired by the non-Indo-European languages, Finnish and Hungarian, but with some influence from Sanskrit. For the Kingdom’s religious beliefs and its ancient history, I called on Robert Graves’ interpretation of the Greek myths – that they reflect a patriarchal religion and society imposed on an ancient matriarchy. Sometime ago I spent several years as a practising Tibetan Buddhist, and much of what I learned during that time about Tibet’s geography, social structure and history has gone into creating Kiralussats as an isolated mountain kingdom ruled by a reincarnated God-King and dominated by a powerful monastic clergy. The following stories mark the start of that process. However, since it would be an imaginary place, I would have to do some world-building before I could introduce the young pretender onto the scene. Since reading The Daughter of Time by Josephine Tey, I have been intrigued by the mystery of the Princes in the Tower, especially after learning about Perkin Warbeck (see Perkin Warbeck: the Man who would be King) the young pretender who claimed to be Richard, Duke of York, the younger of the two princes.Īt one stage I considered writing a novel about Perkin Warbeck, but then I realised that it had been done and done – as an historical novel, that is – but what if, I thought, I set a story inspired by Perkin Warbeck in an imaginary world, a world that is different, but not so different, to our own. You can easy walk Death March just about with any build. Just because you don't see value in something, doesn't mean there is none. I DO NOT say that's your build is bad, I just say it's not true to say that other builds are not viable. Hardly something you'd not want and another instance of where you have contradicted yourself. More of your ignorance, if you want to go for as high DPS as possible, the tier 4 oil skill will help you deal 100% more critical damage when your adrenaline points are at max. Moreover, what's to keep me from dressing up in cat school armor and get the tecnique and rack things up even more but still have the added benefit of high survival and resilence the alchemy tree provides? Again, who gets adrenaline points faster? Your nine skills = 5*8 = 40+30 from razor focus = 70%. Again what keeps you from adding Razor focus and that ability that eliminates adrenaline loss from taking damage? In fact those two skills are exactly what I have in my build. Ursine gear comes with an inherit boost to adrenaline gain +70% in total (with swords), so 6 combat skills = 5*6 = 30 + 70 = 100%. Some people want to go signs or alchemy for the idea of offensive gains, not defensive ones.Įxtra crit damage only applies to the sword base damage, it's not as big as you think and 20% more damage is of course cool, but it's not even close to being "no where near your damage" Anyone can do Death March with ease with a max DPS build, so if you're going signs or alchemy it's for wanting to roleplay a tank, not because it's optimal for kill times.Īlchemy and Sign Builds should have had more DPS aspects fused into its utilities. You forgot Battle Trance and Razor Focus, which increase your adrenaline gain substantially and keep those adrenaline points there, which means your ramp up to the 30% attack power is vastly reduced, so you are doing damage with maxed adrenaline bonus much earlier.Īlchemy and signs are utility trees, and hardly necessary, which the OP rightly complained about. I can't even trust you anymore given how you wave away substantial numbers. It's a DPS gain.Īnd 20% extra damage and 100% extra crit damage from cat school techniques, how is that not an immense DPS boost. Even if you get 30-40% sign intensity total from slotting the 3 signs on a greater blue mutagen, you are debuffing armor by that amount. Um, you forgot the combat skill that increases enemy damage taken by 25% upon being hit by a heavy attack (so you heavy attack a single time purely to apply that debuff_.
